Every skin penetration study used in regulatory skin penetration testing is initiated with an assessment of the integrity of the skin to be used.  There are many methods to assess skin barrier function including tritiated water penetration, electrical resistance and transepidermal water loss (TEWL).  A barrier function acceptace criterion must be determined for the different types of equipment (flow and static diffusion cells) as well as exposure area (0.64 and 3.14 cm2).  This project will evaluate a large body of historical data generated within this laboratory to identify new critical cut of values.  Where necessary, additional data points will be created.

Success will be the creation of a robust method to replace our established method.  We would also aim to publish a poster in a scientific meeting.

The Department of Metabolism and Pharmacokinetics provides support to the pharmaceutical, veterinary pharmaceutical and chemical/agrochemical industries by evaluating the metabolism and disposition of new chemical and biological entities in vivo.  A key test method to help us achieve this is Quantitative Whole Body Autoradiography (QWBA).  When a radioactive-labelled drug is administered in vivo, this technique allows us to generate images of a whole biological system to assess exactly which tissues the drug is distributed to. This gives us valuable information as to whether the drug is going to desirable or undesirable areas of the body, and how long it stays there.

Human blood is currently used as a vehicle for radioactive standard controls as part of the QWBA technique. Blood provides an excellent comparison as a biological matrix to the test systems under analysis, however its supply relies on healthy volunteers and a phlebotomist, meaning that it is not instantly replenishable by the testing laboratory. The blood also provides a challenge during preparation due to blood clots and is time-consuming to prepare.

The aim of this project is to perform a comparative study to investigate the potential of a common emulsifier/thickener called carboxymethylcellulose (CMC) to replace human blood samples as a vehicle for radioactive control standards in QWBA tests.

Scottish salmon, produced by aquaculture, is the UK’s largest single food export. Salmon farmed in the sea, like any other animal species, attract parasites. One prevalent parasite is sea lice which are managed by a range of methods, both preventative and via interventions or treatments to remove them from adhering to the salmon’s skin. When a medicine is used in the open sea environment it is strictly controlled and monitored by the Scottish Environmental Protection Agency. There is a substantial body of publicly available data on medicine use in aquaculture which awaits analysis.

The Scottish Aquaculture Innovation Centre wishes to carry out a short project to examine the use of medicines over time. This is a novel data analysis exercise and would provide a candidate with experience handling large volumes of information.

The project will look at the implementation of a syringe pump to deliver a constant virus spike to a viral filtration load in a continuous manner allowing the operation of a coupled pre filter and virus filter. The initial phase will be familiarisation with the set up and operation of the kit and use the syringe pump to evaluate spiking at a selcted flow rates spannign the typical range observed during viral filtration operation. The initial studies will use a saline solution and the spike ration evaluated by conductivity.

Phase 2 will evaluate a minimum of 2 flow rates using a virus spike, spiking into an appropriate media.  Consistency of spike addition will be assessed by measurment of viral titre in selected samples, taken suring the run, using a TCID50 infectivity assay.

Phase 3 will look at implementing inline spiking in situ i.e. during viral filtration (run in coupled mode) comparing to batch spiking and decoupled filters.  If time permits an evaluation operating both a pump and compressed gas driven viral reduction filtration system will be evaluated.
